    ATCC her2 negative mcf7
EMT and <t>HER2</t> silencing by biphasic metaplastic carcinoma. A A typical case of metaplastic carcinoma shows a biphasic epithelial (E) and mesenchymal pattern (M). B Strong CDH1 expression is present in the former (E) whereas CDH1 expression is silenced in the latter (M). C HER2 gene amplification (red/green signal ratio > 10X) by FISH is present equally in both areas. D But high HER2 expression present in the epithelial phase ( E ) is silenced in the mesenchymal phase (M). Scale bars are provided

Image analysis and EMT comparisons in IDC v MC. A TMAs were subjected to ERAs and SRAs designed to identify monophasic non-embolic and embolic regions of infiltrating IDC and biphasic epithelial and mesenchymal regions of MC. 100 regions of interest from each of these areas were selected and studied for CDH1 and VIM immunoreactivities. SRAs showed the relative signal intensities of each of these proteins in the indicated areas of IDC and MC. For each of these parameters, the graph depicts calculated means ± SD. Differences of significance are depicted. B In emboli v non-emboli of IDC, relative mRNA levels compared to reference levels in human dermal fibroblasts (HDF) showed high steady state CDH1 and low EMT-related genes (TWIST1, VIM and FN1) equally in both emboli and non-emboli. In biphasic MC, the mesenchymal phase (MC-mesen) compared with the epithelial phase (MC-epith) showed significant EMT. In cases of HER2 overexpressing IDC and MC, overexpression of HER2 persisted in the emboli of IDC but was dramatically decreased in the mesenchymal regions of MC which had undergone EMT. Results depicted are mean + standard deviation of 5 separate experiments. Each experiment also had five separate replicates

    Induced EMT and HER2 silencing. A MCF7 grows as epithelial monolayer clusters which are CDH1 positive ( B ) and VIM ( C ) negative, HER2 unamplified by FISH ratio of red/green signals < 4 ( D ) and negative for HER2 expression ( E ). TGFβ1 induces EMT manifested by induction of spindle cell morphology ( F ) and negative CDH1 immunoreactivity ( G ) and positive VIM immunoreactivity ( H ). HER2 remains unamplified by FISH ( I ) and HER2 expression remains negative ( J ). HTB20 similarly grows as epithelial monolayer clusters ( K ) which are CDH1 positive ( L ) and VIM ( M ) negative but exhibit HER2 gene amplification by FISH ratio of red/green signals > 4 ( N ) and resulting HER2 overexpression ( O ). TGFβ1 induces EMT manifested by induction of spindle cell morphology ( P ) and negative CDH1 immunoreactivity ( Q ) and positive VIM immunoreactivity ( R ). HER2 in HTB20 remains amplified by FISH ( S ) but TGFβ1 completely silences HER2 expression ( T ). Scale bars are provided

Induced EMT transcription in breast carcinoma cell lines. In the MCF7 line, TGFβ1 induced EMT. In the HTB20 line overexpressing HER2, TGFβ1 both induced EMT and silenced HER2 expression. With both cell lines, relative mRNA levels compared to reference levels in human dermal fibroblasts (HDF) showed significant decreases in CDH1 mRNA levels and similarly significant increases in EMT-related gene mRNA levels (TWIST1, VIM and FN1).
